Metachromatic Leukodystrophy (MLD) is a rare genetic disorder that affects the nervous system. It is caused by mutations that lead to a deficiency in an enzyme called arylsulfatase A (ARSA). This enzyme deficiency results in the accumulation of certain fats in the brain and other parts of the body, leading to progressive damage to the nervous system. The study focuses on a specific form of MLD known as Late Juvenile Metachromatic Leukodystrophy, which typically begins in children between the ages of 7 and 17.
The purpose of this clinical trial is to evaluate the safety and effectiveness of a gene therapy treatment called OTL-200. This treatment involves a single infusion of a product named Libmeldy, which contains a substance known as atidarsagene autotemcel. This therapy aims to introduce a healthy copy of the ARSA gene into the patient's cells, potentially correcting the enzyme deficiency. The treatment is administered through an intravenous infusion, meaning it is delivered directly into the bloodstream.
Participants in the study will receive the OTL-200 infusion and will be monitored over a period of time to assess changes in ARSA activity levels in the cerebrospinal fluid (CSF) and the brain. The study will also track other health indicators to determine the treatment's impact on the progression of the disease. The trial is open-label, meaning both the researchers and participants know what treatment is being administered, and it is non-randomized, so all participants receive the same treatment. The study aims to provide valuable information on the potential benefits of this gene therapy for individuals with Late Juvenile Metachromatic Leukodystrophy.
